Durch die intranasale Verabreichung könnte das Medikament über die Riechlappen in das zentrale Nervensystem (ZNS) gelangen, wodurch die Blut-Hirn-Schranke (BHS) umgangen wird, was den First-Pass-Metabolismus umgeht und folglich die Bioverfügbarkeit des Medikaments erhöht. ZMT-Niosomen wurden mithilfe der Lipidfilm-Hydratisierungstechnik hergestellt. Die optimierte niosomale Formulierung wurde für die Herstellung einer in situ Gelformulierung verwendet. Die entwickelten Formulierungen wurden im Hinblick auf Vesikelgröße, Form, Zeta-Potential, Einschlusseffizienz, Wirkstoffgehalt und In-vitro-Diffusionsstudie, Mukoadhäsionsstärke, Permeationsstudie, FTIR-, DSC- und XRD-Studien charakterisiert. Die FTIR- und DSC-Studien ergaben, dass es keine chemische Wechselwirkung zwischen dem Arzneimittel und den Hilfsstoffen gab. Die ZMT-Niosomen wiesen Partikelgröße, PDI, Zeta-Potential, prozentuale Einschlusseffizienz und Arzneimittelgehalt auf: 149 nm, 0,223, -28,9, 88,16±0,8 % bzw. 96,23±1,2 %. Die In-vitro-Diffusionsstudie der Niosomen zeigt 96,23±0,7% nach 8 Stunden. Die Permeationsrate des in situ Niosomengels und des reinen Arzneimittels lag bei 98,56 % und 79,46 %.
